Recent scientific studies have examined sildenafil's effects on cancer cells, particularly in colorectal cancer, where researchers have identified mechanisms suggesting the drug may interfere with cholesterol metabolism and cellular processes involved in tumor growth and metastasis. One recently published study found that PDE5 inhibitors such as sildenafil disrupted cholesterol trafficking within cancer cells by affecting the NPC1 cholesterol transport pathway, reducing the availability of cholesterol needed for cancer cell migration and metastasis. Researchers also observed that combining sildenafil with statins produced additive anti-metastatic effects in preclinical models, though they emphasized that additional clinical research is needed before these findings could translate into standard patient care. Earlier laboratory research likewise demonstrated that sildenafil inhibited the growth of human colorectal cancer cells in both cell culture and animal models by promoting cell-cycle arrest and programmed cell death.
